白花蛇舌草水提部位体内外抗肿瘤实验研究 被引量:35

摘要 目的探讨白花蛇舌草水提部位(HD-1)体内外抗肿瘤活性。方法体外实验采用MTT法检测HD-1对人肝癌细胞HepG2、人直结肠癌细胞株HCT-116的生长抑制作用;体内实验采用S180实体瘤小鼠动物模型评价HD-1不同剂量(100,50 mg.kg-1)对S180肉瘤的生长抑制作用及对荷瘤小鼠体重、抑瘤率、胸腺指数、脾指数等的影响。结果 HD-1在体外可显著抑制HepG2、HCT-116肿瘤细胞的增值,其效果与剂量和作用时间相关;体内试验表明,与模型组比较,HD-1高、中剂量组(100,50 mg.kg-1)显著抑制S180肉瘤生长,平均抑制率分别为12.5%和33.1%,脾指数显著升高(P<0.05);环磷酰胺(CTX)联合HD-1中剂量组与CTX(20mg.kg-1)组比较,联合组胸腺指数、脾脏指数、肝指数显著提高(P<0.05),肺指数显著降低(P<0.01)。结论 HD-1在体外能明显抑制HepG2、HCT-116细胞的增值,体内能抑制荷瘤小鼠S180肉瘤生长。 Objective To study the in-vitro and in-vivo antitumor effect of water extract(HD-1 ) from Herba Hedyotis Diffusae. Methods In in-vitro experiment, MTT assay was applied to detect the inhibitory effect of HD-1 on the growth of human hepatic cancer HepG2 cell line and human colon carcinoma HCT-116 cell line. In in-vivo experiment, the mice models of S180 solid tumor were established to evaluate the effect of different concentrations of HD-1 (100, 50 mg kg-1) on inhibiting S180 solid tumor growth, and on body weight , spleen index, thymus index , tumor mass weight and the rates of tumor inhibition in tumor-bearing mice. Results HD-1 could markedly inhibit HepG2 and HCT-116 cell proliferation in-vitro, and the effect was in dose- and time-dependant manner. The results of in- vivo experiment showed that tumor growth was apparently inhibited by HD-1 (100, 50 mg kg-1) with inhibitory rate being 12.5 % and 33.1%, respectively; spleen index was significantly increased in HD-1 groups as compared with that in model control group(P 〈 0.05). When used together with CTX(20 mg kg-1), HD-I(50 mg kg-1) in- creased the spleen index, thymus index and liver index (P 〈 0.05 ), and decreased the lung index significantly (P 〈 0.01). Conclusion HD-1 exerts an obvious effect on inhibiting HepG2 and HCT-116 cell proliferation in vitro, and on inhibiting the growth of sarcoma-180 in vivo in mice.
