河南省2010年肠道病毒EV71型分离株全基因组序列分析 被引量:4

Complete sequence analysis of Enterovirus 71 strain from a fatal case of hand-foot-mouth disease during an epidemic of Henan province in 2010

摘要 目的:了解2010年河南省流行的肠道病毒(EV)71型基因特征。方法:筛选2010年河南省分离的EV71型病毒株1株(EV71/Henan/DC/2010),进行全基因组序列测定和基因进化特性分析。结果:EV71/Henan/DC/2010与其他EV71病毒株相比,编码区无核苷酸的缺失或插入,5’UTR区和3’UTR区长度和序列有一定差异。核苷酸同源性比较显示,EV71/Henan/DC/2010与2008年北京暴发流行株同源性最高(99.2%),与安徽阜阳暴发流行株和河南省暴发流行株均具有很高的同源性(98.1%和99.1%);与EV71各亚型代表株的比较结果显示,除VP3区以外,EV71/Henan/DC/2010各区均与C4亚型代表株同源性最高;氨基酸同源性比较结果显示,EV71/Henan/DC/2010大多数核苷酸变异属无义突变,相应氨基酸并未随之发生改变。根据VP1区基因序列构建遗传进化分析树显示,EV71/Henan/DC/2010与C4亚型同属一个分支。结论:EV71/Henan/DC/2010与2008年北京EV71流行株亲缘关系最为接近,同属于C4亚型。与我省及我国既往流行毒株相比,未发生大的变异。 Aim: To investigate the genetic characteristics of Enterovirus 71(EV71) circulating strains of Henan province in 2010.Methods:The EV71 strain isolated from a fatal case of hand-foot-mouth disease during an epidemic of Henan in 2010 was chosen.Its complete genome was sequenced and analyzed comparatively.Results: The results showed that EV71/Henan/DC/2010 had no insertion or deletion detected in the coding region but several insertions and deletions in 5’and 3’UTR.Phylogenetic analysis of EV71/Henan/DC/2010 and reference strains showed it had the highest nuclyeotide homology with 2008 Beijing epidemicstrain FJ606448(99.2%),and with Anhui epidemicstrain EU703813 and Henan epidemicstrain GU366191(98.1% and 99.1%).The majority nucleotide variations were nonsensemutation,and the corresponding amino acid did not change along with it.In all but VP3 regions,EV71/Henan/DC/2010 had the highest nucleotide homology with C4 subtype.It clustered with reference strains of C4 subtype in the phylogenetic tree.Conclusion: EV71/Henan/DC/2010 strain has the nearest genetic relationship with 2008 Beijing epidemicstrain FJ606448,both belong to the C4 subtype.EV71/Henan/DC/2010 strain has not remarkable variation.
