摘要
目的:探讨两头毛水提物(IAR-Water)的抗氧化活性及抗氧化机制。方法:72只小鼠分成6组,每组12只,分别为空白组,模型组,阳性药物维生素E组(200mg/kg),IAR-Water低、中和高剂量(分别相当于药材1.9、3.8和7.6g/kg)组,给药同时,模型组、阳性药物维生素E组、IAR-Water低、中和高剂量组小鼠每天颈背部皮下注射D-半乳糖1g/(kg·d)建立衰老小鼠模型,空白组皮下注射等体积注射用水。7周后对小鼠心、肝和肾组织中丙二醛(MDA)、超氧化物歧化酶(SOD)和还原型谷胱甘肽(GSH)的含量进行测定。结果:各组小鼠心、肝、肾中MDA、SOD和GSH的含量比较,差异有统计学意义(P<0.05)。与模型组比较,阳性药物维生素E组和IAR-Water中和高剂量组心、肝、肾组织中MDA的含量下降,SOD和GSH的含量升高(P<0.05)。结论:IAR-Water可能通过清除羟自由基而发挥抗氧化作用。
Aim: To study the antioxidative activity and its mechanism of the water extract from incarvillea arguta royle(IAR).Methods:A total of 72 mice were randomly divided into six groups(12 mice in each group):the normal group,model group,positive medicine vitamin E group (200 mg/kg),IAR-Water low,medium,high dose group (equivalent drugs 1.9,3.8,7.6 g/kg).At the same time of administration,the model group,positive control group and reagent mice were injected D-galactose 1g/(kg·d) by SC,the normal group was injected the same volume of water.Seven weeks after administration,the levels of malondialdehyde(MDA),superoxide dismutase(SOD),and glutathione peroxidase(GSH) in tissues of heart,liver,and kidney were detected.Results:The levels of MDA,SOD,and GSH of 6 groups in tissues of heart,liver,and kidney were obviously different (P〈0.05).The levels of MDA were lower and the levels of SOD and GSH were higher in positive medicine vitamin E group,IAR-Water medium,high dose groups than those in model group(P〈0.05).Conclusion:IAR water extract has the antioxidative activity by eliminating oxygen free radicals.
