摘要
以大岩桐幼嫩叶片、老叶、带腋芽的茎段为外植体,进行试管诱导,并进行继代、生根、移栽试验。结果表明:大岩桐诱导外植体以带腋芽的茎段效果最好;适宜的启动培养基、增殖培养基和生根培养基分别为MS+6-BA 1.0 mg.L-1+NAA 0.2 mg.L-1、MS+6-BA 0.5 mg.L-1+NAA 0.1 mg.L-1和1/2MS+NAA0.5 mg.L-1;用珍珠岩做移栽基质,炼苗成活率可达85%。
The test-tube induction,subculture,rooting and explants transplantation were conducted on explants of Sinningia speciosa including young leaves,adult leaves and stems with axillary bud.The results showed that the stem with axillary bud was the best explant for tissue culture of S.speciosa.The suitable initial medium,multiplication medium and root medium were MS+6-BA 1.0 mg·L-1+NAA 0.2 mg·L-1,MS+6-BA 0.5 mg·L-1+NAA 0.1 mg·L-1 and 1/2MS+NAA0.5 mg·L-1 in this experiment.The survival rate of transplanting seedlings reached to 85% with perlite as transplanting medium.
