摘要
研究了在PBS缓冲介质中,一种检测癌胚抗原的新型免标记电化学免疫传感器的制备及应用,石墨烯与甲苯胺蓝复合物饰于玻碳电极表面,通过循环伏安法对修饰的电极进行表征。基于以[Fe(CN)6]3-/4-为氧化还原探针,癌胚抗原抗体反应引起[Fe(CN)6]3-/4-探针的电流响应的变化,来实现癌胚抗原的检测,癌胚抗原的浓度在0.3~10.0 ng/mL范围内与峰电流呈良好的线性关系,回归方程为Δi=-1.926ρ+0.413,相关系数为0.9903,检测限为0.1 ng/mL,该传感器具有良好的重现性、选择性和稳定性,用于人血清样品的测定获得满意结果。
A novel label-free electrochemical immunosensor for the detection of CEA has been developed by immobilizing anti-CEA on a graphene sheet-Toluidine blue composite modified glassy carbonelectrode.The modified electrode was characterized by cyclic voltammetry.The relative current response to the reaction of antigen and antibody based on the redox probe [Fe(CN)6]3-/4-for the determination of CEA was recorded.The detection is based on the peak current change of the modified electrode before and after the antigen-antibody reaction.The linear range is 0.3~10.0 ng/mL.The linear regression equation is Δi=-1.926c+0.413(r= 0.9903).The detection limit is 0.1 ng/mL.The sensor has good reproducibility,selectivity and stability,and it has been applied to the determination of human serum with satisfactory results.
