β-防御素在金华猪和长白猪肠道中表达规律的研究 被引量:10

摘要 本试验旨在研究金华猪和长白猪从初生到60日龄不同肠段中3种β-防御素[β-防御素-1(pBD-1)、β-防御素-2(pBD-2)和β-防御素-3(pBD-3)]基因表达规律,并比较β-防御素基因在2个品种之间的表达差异。选取初生、20、40和60日龄的金华猪和长白猪各3头,屠宰后取十二指肠、空肠、回肠和结肠组织,实时定量PCR法检测各肠段pBD-1、pBD-2和pBD-3基因的表达量。结果表明:1)从初生到60日龄,随日龄的增加pBD-1和pBD-3基因在猪肠道中的表达量升高,pBD-2基因在十二指肠和空肠中的表达量升高,而在猪结肠和回肠中的表达量先下降后上升。2)在20、40和60日龄时,金华猪各肠段中3种β-防御素基因的表达量均高于长白猪。3)初生和20日龄,金华猪十二指肠pBD-1基因表达量极显著高于长白猪(P<0.01);60日龄pBD-1基因在空肠、回肠和结肠中的表达量,金华猪显著高于长白猪(P<0.05),pBD-2基因在十二指肠、回肠和结肠的表达量,金华猪显著高于长白猪(P<0.05),金华猪pBD-3基因在十二指肠、空肠和回肠的表达量显著高于长白猪(P<0.05),结肠中的表达量极显著高于长白猪(P<0.01)。结果显示,初生到60日龄,pBD-1和pBD-3基因在猪肠道中的表达呈上升趋势,金华猪肠道中3种β-防御素的表达水平在20、40和60日龄均高于长白猪。 The study was conducted to investigate gene expression regularity of the three kinds of β-defensins [β-defensins-1(pBD-1),β-defensins-2(pBD-2) and β-defensins(pBD-3)] in different segments of intestinal tract in Jinhua and Landrace pigs from new-born to 60 days of age,and to compare differences of β-defensins gene expression between the two breeds.Three heads of Jinhua pigs and Landrace pigs(new-born,20,40 and 60 days of age) were selected and slaughtered,respectively,and tissue samples of duodenum,jejunum,ileum and colon were collected for analysis of gene expression levels of pBD-1,pBD-2,pBD-3 by real-time PCR.The results showed as follows:1) with the days of age increasing,pBD-1 and pBD-3 gene expression levels in intestine increased,and pBD-2 gene expression level in duodenum and jejunum increased,while that in ileum and colon tended to firstly decrease and then increase.2) At 20,40 and 60 days of age,gene expression levels of the three β-defensins in different segments of intestine in Jinhua pigs were higher than those in Landrace pigs,respectively.3) gene expression level of pBD-1 in duodenum of Jinhua pigs was significantly higher than that of new-born and 20-day-age Landrace pigs(P〈0.01);at 60 days of age,gene expression level of pBD-1 in jejunum,ileum and colon of Jinhua pigs was significantly higher than that of Landrace pigs(P〈0.05),gene expression level of pBD-2 in duodenum,ileum and colon of Jinhua pigs was significantly higher than that of Landrace pigs(P〈0.05),and gene expression level of pBD-3 in duodenum(P〈0.05),jejunum(P〈0.05),ileum and colin(P〈0.01) of Jinhua pigs was significantly higher than that of Landrace pigs(P〈0.05).These results indicate that the gene expression levels of pBD-1and pBD-3 increase in the intestinal tract of pigs from birth to 60 days of age,and gene expression levels of the three β-defensins in Jinhua pigs are higher than those in Landrace pigs at 20,40 and 60 days of age.
