摘要
目的研究大蒜油对二乙基亚硝胺(N-nitrosodiethylamine,NDEA)诱发大鼠肝癌的抑制作用。方法 60只雄性Wistar大鼠随机分为4组:正常对照组、NDEA模型组、大蒜油低、高剂量处理组,每组15只。NDEA模型组和大蒜油处理组均灌胃NDEA(10 mg/kg),2个大蒜油处理组同时给予不同浓度的大蒜油灌胃(20和40 mg/kg),正常对照组给予等体积的玉米油灌胃。观察大鼠体重、肝重、肝脏系数、癌结节数和癌结节发生率的变化,同是对肝脏进行病理组织学检查;并检测血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)、γ-谷氨酰转肽酶(γ-GT)的活性和甲胎蛋白(AFP)的含量。结果与正常对照组相比,NDEA模型组大鼠体重明显降低,肝重和肝脏系数明显增加,血清ALT、AST、ALP和γ-GT的活性和AFP的含量均明显升高(P<0.01);与NDEA模型组相比,大蒜油处理组大鼠各项指标明显好转(P<0.01或P<0.05)。病理结果显示,与NDEA模型组相比,大蒜油低、高剂量组癌结节数和发生率明显降低,且癌细胞分化程度较高。结论大蒜油对NDEA诱发大鼠肝癌形成具有明显的抑制作用。
Objective To observe the inhibitory effect of garlic oil on hepatocarcinoma induced by N-nitrosodiethylamine(NDEA) in rats.Methods Sixty male wistar rats were randomly divided into normal control group,NDEA model group and two treatment groups.They were gavaged with NDEA(10 mg/kg) in model group and were gavaged with garlic oil(20 or 40 mg/kg) in trentment groups.The rats of normal control group were given the same volume of corn oil.Changes of body weight,liver weight,organ coefficiecot of liver,number and incidence of liver nodules,ALT,AST,ALP,γ-GT and AFP in serum were observed and detected.Results Compared with normal control group,body weights were significantly decreased and liver weights,ratios of liver and body weight,the activities of ALT,AST,ALP,γ-GT and the content of AFP in serum were significantly increased in NDEA model group(P0.01).Compared with NDEA model group,all indicators were significantly improved in treatment groups(P0.05 or P0.01).Histopathological examination showed that garlic oil reduced the number and incidence of liver nodules induced by NDEA and led to a higher degree of cell differentiation.Conclusion Garlic oil can protect liver cells from damage and inhibit the development of hepatocarcinoma induced by NDEA.
