摘要
目的研究鸡矢藤环烯醚萜苷(IGPS)对尿酸性肾病(UAN)大鼠的防治作用及其作用机制。方法采用腺嘌呤(100 mg/kg,灌胃给药,qd×28 d)和氧嗪酸钾(250 mg/kg,腹腔注射,每周1次,连续4周)制备UAN大鼠模型,造模4周后,检测各组大鼠血尿酸、血黄嘌呤氧化酶、血肌酐、尿素氮、尿尿酸及肾指数。结果 IGPS(70、140、280 mg/kg,灌胃给药,qd×21 d)可明显改善UAN大鼠的一般症状,明显抑制血清黄嘌呤氧化酶活性、降低血尿酸含量;促进尿尿酸排泄,减少肾组织内尿酸盐沉积物,改善肾功能,降低肾指数。结论 IGPS对腺嘌呤和氧嗪酸钾所致UAN大鼠有较好的防治作用。
Objective To observe the preventive and therapeutic effects of iridiod glycosides from paederia scandens(IGPS) on uric acid nephropathy(UAN),rats and its possible mechanism.Methods The UAN rat model induced by adenine(100 mg/kg,ig,qd×28 d) and potassium oxonate(250 mg/kg,ip,qw×4 wk).After 4 weeks of modeling,the rats in each group of serum uric acid(UA),blood urea nitrogen(BUN),serum creatinine(Cre),xanthine oxidase(XOD),urinary UA and renal indices were examined.Results IGPS(70,140,280 mg/kg,ig,qd×21 d) could significantly improve the UAN in rats in general symptoms,significantly inhibited the serum XOD activity,lower blood UA content;the promotion of urinary UA excretion,reducing kidney tissue urate deposits,improving renal function,lower renal index.Conclusion IGPS has good preventive and therapeutic effect on UAN induced by adenine and potassium oxonate in rats.
