摘要
为了研究二甲苯对小鼠卵巢组织的损伤作用,以健康成年SD雌性小鼠作为实验动物,分3个试验组(低、中、高剂量组)和1个对照组,试验组腹腔注射不同剂量(按体重0.125、0.25、0.5mL/kg)的二甲苯,对照组每天腹腔注射生理盐水。采用石蜡切片和HE染色技术观察二甲苯染毒后小鼠卵巢组织结构的变化;采用分光光度法测定小鼠卵巢组织中超氧化物歧化酶(SOD)的活性和丙二醛(MDA)含量。结果表明,二甲苯染毒导致小鼠卵巢发生萎缩,中、高剂量组的卵泡内都有黑色颗粒,且随二甲苯剂量的增加其病理变化更加明显;中、高剂量组卵巢组织的SOD活性与对照组相比显著下降,而MDA含量显著升高。证实,二甲苯对卵巢组织具有明显的损伤作用,且表现出明显的剂量效应。
In order to study the damage of xylene to the ovary tissue of mouse,healthy adult female SD mice were randomly divided into 3(high,middle,low dose) experimental groups and one control group.The mice in experimental groups were injected different volume of xylene(0.5,0.25,0.125 mL/kg) and the mice in the control group were injected intraperitoneally with normal saline solution every day.Histological structure of murine ovary under different xylene volume were observed by HE staining and paraffin section.The activity of superoxide dismutase(SOD) and content of malondialdehyde(MDA) in the xylene-infected mouse ovary were measured by spectrophotometric method.Results showed that the xylene may lead to ovary atrophy and black solid grains were found in follicle in the middle and high dose group,and pathological changes of ovarian tissue was more obvious with xylene dose increasing.SOD activity were decreased significantly,while the content of MDA was significantly increased in high-and middle-dose groups compared with that of control group.It indicated that xylene may damage the ovary tissue of mice,and showed significant dose effect.
