摘要
目的:研究交泰丸对2型糖尿病大鼠胰岛形态的影响。方法:用高糖高脂饲料喂养大鼠6周,腹腔注射低剂量链脲佐菌素(STZ)37mg/kg制作大鼠2型糖尿病动物模型。给予交泰丸水煎剂灌胃治疗5周,动态观察其治疗后的饮水量、进食量、体重、空腹血糖(FBG)变化,于治疗5周后检测血清游离脂肪酸(FFA)、甘油三酯(TG)、血清胰岛素(INS)水平;并取胰腺在光镜和透射电子显微镜下观察组织形态学变化;免疫组化法观察胰岛素阳性细胞。结果:与模型组比较,交泰丸组在用药1周末即出现FBG降低趋势,5周末有明显差异;3周后进食量明显减少;TG、FFA与模型组比较显著下降;组织形态学观察胰岛结构较完整,细胞数量相对增多,细胞内分泌颗粒较丰富,胰岛周边部导管上皮细胞样细胞增多,并向胰岛中央移行;免疫组化结果显示,交泰丸组胰岛内胰岛素阳性细胞较多,染色较深。结论:交泰丸对2型糖尿病大鼠胰岛结构具有保护作用。
Objective:To observe the influence of jiaotai pill on morphological changes of islets of pancreas in type 2 diabetes mellitus rats.Methods:Models with type 2 diabetes mellitus were made by feeding high-carbonhydrate and high-lipid diet for 6 weeks,than intra-abdominal STZ,experiment was divided into normal,model and jiaotai pill groups,normal and model group were given saline,Chinese group was given jiaotai pill for 5 weeks,dynamic observing the changes of water intake and food intake,body weight and fasting blood-glucose,detecting serum FFA,TG and IND level;taking pancreas tissue to observe the pathomorphology changes by light and electron microscope;insulin positive cells were observed by immunohistochemical method.Results:To compare with model group,FBG of jiaotai pill group began to low after treated 1 week,the difference between jiaotai pillgroup and model group was difference after treated 5 weeks;food intake decreased obviously from the third week,on the fifth weekend,water intake,TG,FFA reduced significantly.The pathomorphology observation showed that the islets structure of jiaotai pill group was more intact than that of model,the number of cells increased,secretory granules of cells were rich,epithelioid cells of islets peripheral ducts increased and migrated to central of islets.The immunohistochemical results in jiaotai pill group showed that the number of insulin positive cells increased,staining got shade.Conclusion:Jiaotai pill can protect the structure of islets of pancreas in type 2 diabetes mellitus rats.
