摘要 将枳和酸橙的实生试管苗上、下胚轴和子叶切割成若干段分别进行离体培养,结果表明,枳的各个切段在添加10^(-4)或5×10^(-5)摩尔BA的MS培养基中,不定芽形成率最高,平均每个外植体不定芽发生总量分别达41个和33.5个,其中以上胚轴发生的不定芽最多,占全部不定芽总量的66.6%。在上、下胚轴中,则以第一切段发生的不定芽最多。添加KT的所有处理,不定芽均较少,形成不定芽最多的处理也仅及N_0B_1处理的四分之一。发根量则以添加10^(-5)摩尔NAA的培养基最多。将枳的不定芽扦插于添加5×10^(-6)或10^(-6)摩尔NAA的MS培养基中,15—20天即可发根成苗。在完全相同的各个处理中,酸橙的不芽定形成量明显少于枳。 Epicotyl,hypocotyl and cotyledon of trifoliate orange and sour orange tubed-seedlings were taken as materials in the experiment and cut into several segments, and cultured in vitro. The results showed that the highest percentage adventitious buds formation occured on MS medium added with 10-4 or 5 × 10-5 mol BA, the total number of mean adventitious buds per trifoliate orange explant was up to 41 and 33.5,respectively;the most adventitious buds were formed on the first epicotyl and hypocotyl. Fewer produced aventitious tuds on MS medium supplemented with KT in all treatments.Percentage rooting was the highest on MS medium containing 10-5 mol NAA. 15-20 days after transferred to MS medium added with 5 × 10-6 or 10-6 mol NAA, the adventitious buds of trifoliate orange rooted and plantlets produced. The number of adventitious buds formed in sour orange was significantly less than that in trifoliate orange in the same treatments.
