摘要 目的:建立高效液相法测定防风中4种色原酮的方法。方法:采用Diamonsil C18(4.6mm×200mm,5μm)色谱柱;流动相:甲醇-水梯度洗脱(甲醇:0~15min,20%~45%;15~25min,45%~70%);流速1mL·min-1。柱温30℃;波长254nm。结果:升麻素苷在0.15~3μg、升麻素在0.02~0.4μg、5-O-甲基维斯阿米醇苷在0.25~5μg、亥茅酚苷在0.01~0.2μg线性关系良好。结论:该方法快速简便,准确可靠,可用于防风中4种色原酮的测定。 Objectiv kovia divaricata. Methods: e: A HPLC method A Diamonsil C18(4. was established for determination of the chromones contents of Saposhni- 6 mm × 200 mm, 5 μm) was used with gradient elution of methanol and water that methanol increased from 20 % to 45 % in 15 minute, and then increased from 45 % to 70 % until 25 minute as mobile phase ; the flow rate was 1.0 mL · min^- 1 at 30℃ and the detector wave length was 254 nm. Results: The relationships between the concentration and the peak area of 4 chromones were good linear relation. The calibration curve of prim-O-glucosylcimifugin, cimifugin, 5-O-methylvisammioside and sec-O-glucosylhamaudol were at the range of 0.15 - 3 μg, 0.02 - 0.4 μg, 0.25 - 5μg and 0.01 - 0.2 μg. Conclusion: The method was rapid, sensitive and accurate, and could be used for the determination of chromones of S. divaricata.
