摘要 目的:应用响应曲面法优化铁皮石斛多糖提取的最佳工艺条件,为应用于生产提供参考。方法:在单因素试验的基础上,采用响应曲面法的中心组合设计,考察分析提取温度、提取时间、提取次数、液料比4个主要因素对石斛多糖提取率的影响,对铁皮石斛多糖的提取工艺参数进行优化分析。结果:铁皮石斛多糖提取的最优条件为:提取温度为100℃、提取时间为2.5h、料液比1∶17.2(m/V)、提取次数为3次。在此条件下,理论计算提取率达到51.12%,实测提取率为51.08%,与模型高度拟合。结论:该工艺便捷、快速、易操作、得率较高,可用于铁皮石斛多糖的提取。 Objective: To obtain the optimal conditions of ultrasonic-assisted extraction of polysaccharides from Dendrobium candidum, and to provide the theoretical foundation for the further processing and utilization of Dendrobi- urn candidum. Methods: On the basis of the single factor test, response surface methodology was applied to optimize the main extraction conditions. Results: The results showed that extraction time, water-feed ratio and extraction tem- perature all had significant effects on the extraction rate of polysaccharides. Obvious correlation was found to be existed between extraction time and extraction temperature. The optimal extraction time was 2.5 h and the optimal water-feed ratio was 17.2, and the optimal extraction temperature was 100 ~C. Under these optimized conditions, the extraction rate of polysaccharides was 51.12 %. Conclusion: Multivariate regression analysis showed that the model of extraction time, water-feed ratio, extraction temperature and extraction rate of polysaccharides could be forecasted by applying the model.
