Determination of tabersonine in the seeds of Voacanga africana by HPLC

摘要 目的采用HPLC测定伏康树籽中柳叶水甘草碱的含量。方法色谱柱为Agilent Extend-C18柱(150 mm×4.6 mm,5μm);柱温30℃;流动相为甲醇-0.1 mol.L-1醋酸铵(pH6)(70∶30);检测波长290 nm;流速1.0 mL.min-1;进样量20μL。结果柳叶水甘草碱的进样量0.1708~5.4656μg与峰面积具有良好的线性关系,回归方程为:Y=7.2697×103X-9.1125(r=0.9995)。伏康树籽中柳叶水甘草碱的含量为2.65%。结论所用方法操作简便、快捷、结果准确。 OBJECTIVE To measure tabersonine in the seeds of Voacanga africana Stapf.by an RP-HPLC method.METHODS Agilent Extend-C18(150 mm×4.6 mm,5 μm)column was adopted.The mobile phase consisted of methanol and 0.1 mol·L-1 ammonium acetate(70:30)with flow rate of 1 mL·min-1,and the detector wavelength was 290 nm.RESULTS The standard curve was linear in the range from 0.1708 to 5.4656 μg for tabersonine(r=0.9995).The content of the sample was 2.65%.CONCLUSION The method is simple,fast and accurate.
