摘要
目的:探讨盐敏感性高血压大鼠大动脉结构和功能的变化。方法:12周龄SD大鼠(23只)被分为假手术组(10只)和醋酸脱氧皮质酮(DOCA)盐高血压组(13只),观察其外周及中心动脉压,脉搏波速度(PWV)、颈总动脉内膜中层厚度(IMT)和胸主动脉中层厚度(MT)的改变,并分析血管壁胶原/弹性蛋白组分的变化。结果:与假手术组相比,造模结束后DOCA高血压大鼠外周收缩压明显升高[(110.69±4.19)mmHg比(166.58±4.28)mmHg,P<0.05],中心收缩血压(cSBP),中心舒张血压(cDBP),中心平均血压(cMBP)亦显著升高(P均<0.01),同时伴有胸主动脉PWV明显升高[(4.81±0.14)m/s比(6.33±0.20)m/s,P<0.01]。DOCA盐高血压大鼠颈动脉IMT[(46.00±4.00)μm比(78.00±4.90)μm]和主动脉中层厚度(MT)[(120.48±7.93)μm比(194.25±5.66)μm]明显增厚(P均<0.01),血管壁胶原含量亦明显升高[(0.87±0.05)mg/cm比(1.00±0.03)mg/cm,P<0.05]。结论:醋酸脱氧皮质酮盐高血压大鼠存在明显的血压升高和动脉僵硬;除血压影响之外,其动脉僵硬度的改变亦与大动脉管壁增厚及胶原含量变化有关。
Objective: To explore changes of large arterial structure and function in salt sensitive hypertensive rats. Methods: A total of 23 12-week--old SD rats were randomly divided into sham operation group (n: 10) and hypertension induced by deoxycortieosterone acetate (DOCA) salt group (n= 13) . Changes of their peripheral and central arterial pressure, pulse wave velocity (PWV), intima-media thickness (IMT) of carotid and thoracic aorta was observed. Changes of components of collagen/elastin on vessel wall were analyzed. Results: Compared with sham operation group, peripheral systolic blood pressure [SBP, (110.69±4. 19) mmHg vs. (166.58±4. Z8) mmHg, P〈0.05] significantly increased, and there were also significant increase in central SBP (cSBP), central diastolic blood pressure (eDBP), central mean blood pressure (eMBP), P〈0.01 all, and PWV of thoracic aorta [ (4.81±0. 14) m/s vs. (6.33±0.20) m/s, P〈0.01] also significantly increased in DOCA rats at the end of modeling. Carotid IMT [ (46.00 ±4.00) μm vs. (78.00±4.90) μm] and aortic media thickness (MT) [ (120.48±7.93) vm vs. (194.25±5.66) μm] significantly thickened (P〈0.01 both) , and collagen concentration on vessel wall [ (0.87±0.05) mg/cm vs. (1.00±0.03) mg/cm, P〈0.05] also significantly increased in DOCA rats. Conclusion: The blood pressure and artery stiffness in DOCA- salt- hypertensive rats significantly increasetly change of arterial stiffness is related with thicken of large arterial wall and change of collagen concentration besides impact from blood pressure.
