黄芪多糖联合丹参酮对心衰大鼠心肌NF-κB通路过度激活的影响

Influence of astragalus polysaccharides combined with tanshinone on over-activation of myocardial NF-κB pathway in rats with heart failure

目的研究黄芪多糖联合丹参酮对心力衰竭(CHF)模型大鼠心肌核因子-κB(NF-κB)通路过度激活的影响。方法以大鼠左冠状动脉结扎术造成心肌梗死后心衰模型。超声心动图观察心功能变化情况,将成模后大鼠分为6组:模型组、黄芪多糖组、丹参酮组、黄芪多糖加丹参酮组、洛伐他汀组、假手术组。光镜下观察心肌病理形态学变化,免疫组化法检查心肌NF-κB的激活,Western blot法检测心肌NF-κB、抑制蛋白IKB及IKB激酶(IKK)蛋白的表达。结果心肌NF-κB被激活,NF-κB的表达明显升高,而IKB及IKK的表达明显降低,应用药物干预后,NF-κB的表达降低,而IKB及IKK的表达升高,差异有显著性(P<0.05)。结论应用益气活血中药对CHF大鼠心肌的心功能损害有一定防治作用,机制可能与抑制NF-κB转导通路过度激活有关。

Objective To study the influence of astragalus polysaccharides （APS） combined with tanshinone （Tan） on over-activation of myocardial natural factor-κB （NF-κB） pathway in rat model of chronic heart failure （CHF）. Methods The model of heart failure after myocardial infarction was established with the ligation of left coronary artery. The changes of heart function was observed by using echocardiography. All rats were divided into the model group, APS group, Tan group, Aps and Tan group, lovastatin group and sham-operation group. The changes of myocardial pathomophorlogy was observed with light microscope, activation of myocardial NF-κB was detected by using immunohistochemistry assay, and the expressions of NF-κB, profilin IκB （IκB） and IκB kinase （Iκκ） were detected by using Western blot method. Results Myocardial NF-κB was activated, the expression of NF-κB increased sig- nificantly, and the expressions of profilin IκB and Iκκ decreased significantly. After the intervention of medicinal, the expression of NF-κB decreased and the expressions of profilin IκB and Iκκ increased （P 〈 0. 05 ）. Conclusion The application of Chinese medicinal with the actions of supplementing qi and activating blood circulation can prevent the injury of myocardial function in rats with heart failure, and the mechanism may be related to the inhibition of the over-activation of NF-κB pathway.
