摘要
目的利用经典高脂饮食兔动脉粥样硬化模型,观察益气解毒活血配伍方人参、大黄、瓜蒌对高敏C反应蛋白(hs-CRP)、细胞间黏附因子-1(ICAM-1)、血管黏附因子-1(VCAM-1)和单核细胞趋化蛋白-1(MCP-1)的动态影响。方法 20只新西兰兔随机分为3组,中药组、模型组均给予高脂饲料喂养6周,6周后中药组及模型组采用球囊拉伤术损伤髂动脉,术后均继续以高脂饮食喂养3周。正常组全程给予普通兔饲料喂养。中药组全程使用人参(0.64 g/kg)、大黄(0.43 g/kg)、瓜蒌(2.14 g/kg)配方灌胃,模型组、正常组全程给予等量兔饮用水灌胃。Hs-CRP、ICAM-1、VCAM-1、MCP-1测定均采用酶联免疫吸附(双抗体夹心)法。结果益气解毒活血复方能够显著降低hs-CRP、ICAM-1、VCAM-1、MCP-1水平,并达到正常组水平甚至更低。结论益气解毒活血方能够明显抑制动脉粥样硬化相关炎性因子表达,可能是通过抑制炎症反应,降低炎症因子hs-CRP、ICAM-1、VCAM-1、MCP-1上升水平起到延缓动脉粥样硬化进程的作用。
Objective To observe the dynamic influences of medicinal combination with the actions of supplementing qi, removing toxicity and activating blood circulation [ Renshen (Radix Ginseng), Dahuang (Radix et Rhizoma Rhei) and Gualou ( Fructus Trichosanthis ) ] on high-sensitivity C-reactive protein ( hs-CRP), intercellular adhesion molecule-1 ( ICAM-1 ), vascular adhesion molecule-1 ( VCAM-1 ) and monocyte chemoattractant protein-1 (MCP-1) based on rabbit model of atherosclerosis with classic high fat diet. Methods All 20 New Zealand rabbits were randomly divided into 3 groups. The Chinese medicinal group and model group were fed high fat diet for 6 weeks, and then given balloon pulling to injure the iliac artery followed by 3-week feeding of high fat diet. The normal group was fed general fodder. The Chinese medicinal group was intragastrically given formula of Reshen (0.64 g/kg), Dahuang (0.43 g/kg) and Gualou (2. 14 g/kg) and the model group and normal group were given equal amount of drinking water. The levels of hs-CRP, ICAM-1, VCAM-1 and MCP-1 were detected by using enzymelinked immunosorbent assay ( double antibody sandwich). Results The formula with the actions of supplementing qi, removing toxicity and activating blood circulation decreased significantly the levels of hs-CRP, ICAM-1, VCAM-1 and MCP-1 to the levels as those in the normal group or even lower. Conclusion The formula of medicinal combination with the actions of supplementing qi, removing toxicity and activating blood circulation can significantly inhibit the expressions of inflammatory factors related to atherosclerosis and delay the progress of atherosclerosis through inhibiting inflammatory response and decreasing the levels of hs-CRP, ICAM-1, VCAM-1 and MCP-1.
