摘要
采用体外实验和SDS-PAGE电泳法对纤维蛋白溶解酶SPFE-I的溶栓活性、抗凝活性及纤溶机理进行了研究。体外实验证明,该酶终浓度为0.12U/mL时具有体外抗凝血作用,血块在1.5U/mL酶液中的溶解率为72%;采用SDS-PAGE电泳法研究发现,该酶可以水解纤维蛋白原的β、γ链,激活纤维蛋白溶酶原形成纤维蛋白溶解酶,并降解纤维蛋白溶解酶原激活剂抑制剂-1。
Thrombolysis activity,anticoaguiation activity and fibrinolytic mechanism of SPFE-I were studied by vivo experiment and SDS-PAGE. As a result,the enzyme had the function of anticoagulation in 0.12U/mL, and dissolving rate of blood clots in 1.5U/mL was 72% ;β and y-chains of fibrinogen were cut off by SPFE-I, plasminogen could be activted to from plasmin and plasminogen activator inhibitor- I was also able to be degraded by it.
