摘要
目的:研究金钮扣不同提取物的止咳、化痰作用。方法:金钮扣用95%乙醇回流提取得到醇提取物,加水溶解后依次用石油醚、氯仿、乙酸乙酯、正丁醇萃取,浓缩得相应的萃取物和水相溶解部位。采用豚鼠枸橼酸引咳法、小鼠气管酚红法,观察金钮扣不同提取物的止咳、化痰作用。结果:金钮扣醇提取物组、乙酸乙酯组、正丁醇组和空白对照组的咳嗽潜伏期分别为133.5、138.4、137.1、87.8s,咳嗽次数分别为7、8、8和16次,酚红分泌量分别为1.031、1.014、1.262、0.656μg;醇提取物组、乙酸乙酯组和正丁醇组的咳嗽潜伏期延长、咳嗽次数减少、酚红分泌量增加,与空白对照组比较均具有显著性差异(P<0.01或P<0.05)。石油醚组、氯仿组和水溶部分组的咳嗽潜伏期、咳嗽次数及酚红分泌量,与空白对照组比较均无显著性差异。结论:金钮扣醇提取物止咳和化痰作用的有效成分主要集中在乙酸乙酯和正丁醇萃取部位。
OBJECTIVE : To observe the effects of different extracts of Spilanthes paniculata on relieving cough and reducing phlegm. METHODS:S. paniculata was extracted by 95 % alcohol and dissolved in water, and then extracted by petroleum ether, chloroform, ethyl acetate, n-butanol, respectively. Corresponding extracts and aqueous solution parts were obtained. Citric acid-induced cough in guinea pig method and phenol red excretion of mice trachea were use to observe the effects of S. paniculata on relieving cough and reducing phlegm. RESULTS: The cough incubation periods of S. paniculataal acohol extract group, ethyl acetate group, n-butanol group and blank group were 133.5 s, 138.4 s, 137.1 s and 87.8 s, respectively. Cough frequency in those groups were 7 times, 8 times, 8 times and 16 times; phenol red secretion were 1.031 μg, 1.014μg, 1.262μg and 0.656 μg, respectively; In alcohol extract group, ethyl acetate group and n-butanol group, cough incubation period could be extended to reduce cough frequency and increase phenol red secretion. Compared with blank group, there were significant differences(P〈0.01 or P〈0.05). In petroleum ether groups, chloroform group and water soluble part group, latency of cough, cough frequency and the amount of phenol red secretion, compared with blank group, had no significant difference. CONCLUSION: The active ingredients of alcohol extract of S. paniculata on relieving cough and reducing phlegm mainly concentrate in parts of ethyl acetate and n-butanol.
