摘要
目的:研究二甲双胍与消渴热清加味丸合用对糖尿病模型大鼠周围神经病变的改善作用。方法:实验分为6组,即正常对照(等容生理盐水)、模型(等容生理盐水)、甲钴胺(0.275mg·kg-1)、二甲双胍(0.28g·kg-1)、消渴热清加味丸(15.0g·kg-1)、二甲双胍+消渴热清加味丸(0.14g·kg-1+15.0g·kg-1)组。ig给药,连续4周,每2周1次监测大鼠体重、血糖。4周时测定大鼠24h进食量、饮水量、排尿量、尿糖含量、痛觉传导时间、坐骨神经传导速度和血浆中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性以及血栓素B2(TXB2)、6-酮-前列腺素F1α(6-keto-PGF1α)水平。结果:与模型组比较,各给药组大鼠体重无显著性差异,而进食量、饮水量、排尿量、血糖、尿糖含量显著减少,痛觉传导时间显著缩短,坐骨神经传导速度显著增加、SOD活性显著增强、TXB2和6-keto-PGF1α水平显著降低(P<0.01或P<0.05),GSH-Px活性无显著改变;且中西药合用较单独给予二甲双胍在坐骨神经传导速度、痛觉传导时间、血浆中SOD活力和6-keto-PGF1α水平方面效果更好。结论:中西药合用在改善糖尿病大鼠周围神经病变方面优于单独使用二甲双胍,二者表现出明显的协同作用,且能在剂量上降低二甲双胍的用量,减轻二甲双胍的副作用。
OBJECTIVE: To study the improvement effect of metformin combined with Xiaoke reqing jiawei pills on diabetic model rats. METHODS: Wistar rats were randomly divided into 6 groups:normal control group(NS), model group(NS), mecobalamine group (0.275 mg·kg^-1), metformin group (0.28 g·kg^-1), Xiaoke reqing jiawei pills group ( 15.0 g. kg-1) and metformin combined with Xiaoke reqing jiawei pills group(0.14 g·kg^-1+15.0 g·kg^-1). All the animals were administered four weeks intragastrically. The body weight and blood glucose of rats were detected once two weeks. The feeding quantity, potable water volume, voided volume and urine glucose level were detected in the fourth week. The time of pain conduction and sciatic nerve conduction velocity were also detected. The levels of SOD, GSH-Px activity, TXB2, and 6-keto-PGF1α were also detected. RESULTS: Compared with model group, the change of weight was not significant, feeding quantity, potable water volume, voided volume, blood glucose and urine glucose levels of every treatment group decreased significantly, the time of pain conduction shortened, sciatic nerve conduction velocity increased while SOD activity increased and the contents of TXB2 and 6-keto-PGF1α decreased significantly. GSH-Px activity had no significant change. Western medicine combined with traditional Chinese medicine was better than metformin in above index. CONCLUSION: Drug combination is better than metformin alone in improving peripheral neuropathy in diabetic rats. There is a synergistic effect when model rats are treated with the metformin combined with improved Xiaoke reqing jiawei pills. Besides, the dosage of metformin is reduced and the toxic and side effects from metformin could be lowered.
