摘要
[目的]探究白子菜组织培养最佳培养基配方实现其快速繁殖。[方法]研究不同培养基配方对白子菜组织培养过程中愈伤组织的产生、芽和根分化效果的影响。[结果]④MS+BA 1.5 mg/L+NAA 1.2 mg/L+蔗糖30 g/L+琼脂粉6 g/L为白子菜形成愈伤组织的最佳培养基;⑤花宝一号+BA 0.1 mg/L+NAA 0.4 mg/L+蔗糖30 g/L+琼脂粉6 g/L为诱导愈伤组织形成芽的最佳培养基;瑏瑢MS+BA 1.5 mg/L+NAA 1.2 mg/L+蔗糖20 g/L+活性炭1 g/L+琼脂粉6 g/L为最佳生根培养基。[结论]活性炭对白子菜组织培养有影响,具有抑制愈伤组织形成和促进生根的作用。
[Objective] The aim was to explore the best culture medium formula of tissue culture of Gynura divaricata(Linn.) DC..[Method] The effects of different culture medium formula on callus initiation,differentiation of bud and root were studied.[Results] MS+BA 1.5 mg/L+NAA 1.2 mg/L+saccharose 30 g/L+gelose 6 g/L was the best culture medium of callus initiation of Gynura divaricata;HuaBao No.1+BA 0.1 mg/L+NAA 0.4 mg/L+saccharose 30 g/L gelose 6 g/Lwas the best culture medium which made callus form bud;MS+BA 1.5 mg/L+NAA 1.2 mg/L+saccharose 20 g/L+acitve carbon 1 g/L+geolose 6 g/L was the best rooting medium.[Conclusion] Active carbon affected the propagation speed of culture medium of Gynura divaricata -- restraining callus initiation and accelerating the radication.
