胃癌前病变大鼠中抑癌基因APC、P16表达与浊毒的相关性研究

Correlation Between Expressions of Tumor Suppressor Genes APC,P16 in Rats with Precancerous Lesions and Turbidity Toxin

目的:观察化浊解毒中药对胃癌前病变模型大鼠胃黏膜APC、P16蛋白表达的影响,探讨胃癌前病变大鼠中抑癌基因APC、P16的表达与浊毒之间所存在的相关性。方法:采用结合致癌物的综合造模方法复制胃癌前病变大鼠模型,将造模大鼠随机分为化浊解毒中药组(以下简称"中药组")、模型对照组、空白对照组3组。造模成功后中药组给予化浊解毒中药灌服,模型对照组和空白对照组均给予生理盐水灌胃。连续给药4周后将大鼠断头处死,解剖取胃,石蜡包埋连续切片,用免疫组化方法检测胃黏膜APC和P16蛋白的表达。结果:与空白对照组相比,模型组大鼠胃黏膜APC和P16蛋白的表达明显降低(P<0.05)。与模型对照组比较,中药组大鼠胃黏膜APC和P16蛋白的表达明显升高(P<0.05)。结论:化浊解毒中药能上调胃癌前病变大鼠抑癌基因APC、P16的水平,增加APC、P16蛋白的表达,从而抑制细胞增殖和诱导细胞凋亡,说明胃癌前病变大鼠中抑癌基因APC、P16表达与浊毒之间存在相关性。

Objective： To observe the effects of Chinese herbs for resolving turbidity and toxin on APC,P16 protein expression in rats with gastric precancerous lesions,and to probe into the correlation between expressions of tumor suppressor genes APC,P16 and turbidity toxin.Methods： The comprehensive modeling approach plus carcinogens was used to copy the model rats with gastric precancerous lesions,the rats were randomly divided into three groups： Chinese medical group,model control group and blank control group.Chinese medical group was fed with Chinese herbs for resolving turbidity and toxin,while model control group and blank control group were given normal saline.The rats were decapitated and dissected after four weeks of continuous administration to take the stomach,and to paraffin-embed the serial sections,and then the immunohistochemistry was used to detect the gastric mucosal APC and P16 protein expression.Results： Compared with the blank control group,the gastric mucosal APC and P16 protein expression of the model control group was significantly lowered（P0.05）.Compared with the model control group,the gastric mucosal APC and P16 protein expressions of the Chinese medical group was significantly increased（P0.05）.Conclusion： Chinese herbs for resolving turbidity and toxin can increase the expression levels of tumor suppressor genes APC,P16 in rats with gastric precancerous lesions,and increase APC,P16 protein expressions,thereby inhibiting cell proliferation and inducing apoptosis,which indicates the correlation between expressions of tumor suppressor genes APC,P16 and turbidity toxin.
