胆囊癌及胆囊炎性组织细胞膜的傅里叶变换红外光谱技术研究 被引量:2

摘要 目的研究胆囊癌及胆囊炎性组织细胞膜上生物大分子蛋白质及脂质的红外光谱改变。方法应用傅里叶变换红外光谱技术(FTIR)对胆囊癌和炎性组织的细胞膜进行检测,并分析其蛋白质和脂质的红外变化。结果胆囊癌和炎性组织细胞膜上生物大分子蛋白质和脂质的红外光谱存在明显差异:癌组织中酰胺Ⅱ带较弱、峰型低平,酰胺Ⅰ带较强、峰型宽大,相对吸收强度Ⅱ650/Ⅱ550明显升高;癌组织中与脂质相关的吸收谱带出现概率高,且峰型高尖,相对吸收强度Ⅰ2957/Ⅱ650和Ⅱ740/Ⅱ400的比值增加;相对吸收强度Ⅰ1460/Ⅱ400的比值在正常组织细胞膜中较高。结论胆囊炎性与癌组织细胞膜上大分子红外光谱存在不同,通过FTIR能对细胞癌变早期生物大分子改变做出客观反映。 Objective It is to observe the changes of protein and lipid in membrane of gallbladder cancer and cholecystitis by fourier transform infrared spectroscopy(FTIR).Methods The membranes of gallbladder cancer and cholecystitis tissue samples were detected by FTIR and data were summarized by Omnic 5.0 software.Results There was significant difference in FTIR spectral between cholecystitis tissue and cancerous membrane.In membrane of gallbladder cancer,amide Ⅱ was weaker and lower,amide I was stronger and higher;the ratio of Ⅱ650/Ⅱ550 was raised.In membrane of gallbladder cancer,2957,2927,2853 and 1740 cm-1 were stronger and higher,the ratios of I2950/II650 and Ⅱ740/Ⅱ400 were raised respectively.The ratio of Ⅱ460/Ⅱ400 in membrane of cholecystitis tissue was higher than that in gallbladder cancer tissue.Conclusion There are significant different in FTIR spectral in membrane of cancer tissue and cholecystitis,RTIR can early and objectively reflect the changes of biological macromolecules in the cancerous cells.
