摘要
目的探讨抑癌基因PTEN蛋白在胆囊癌及癌旁正常组织中的表达,及其与微血管密度(MVD)和临床病理特征的关系。方法采用免疫组化SP法对29例胆囊腺癌、8例胆囊鳞癌及其相应癌旁组织中PTEN蛋白进行检测,计算微血管密度,分析PTEN在不同组织中的表达与MVD和胆囊癌组织类型、分化程度等的关系。结果胆囊癌组织中PTEN蛋白阳性表达率低于癌旁正常胆囊黏膜组织(P<0.01),MVD高于癌旁正常组织(t=-7.941,P<0.01);胆囊癌组织中PTEN蛋白阳性表达与患者性别、年龄、组织类型及有无淋巴结转移无明显相关性,但与组织分化程度明显相关。高分化胆囊癌组MVD显著低于低分化组(t=3.275,P<0.01),淋巴结转移组MVD值显著高于无淋巴结转移组(t=-2.931,P<0.01)。结论 PTEN蛋白在胆囊癌组织中阳性表达率较低,PTEN基因突变或缺失可能促进肿瘤血管形成,与肿瘤的生长、分化等相关。该基因在胆囊癌发生发展中可能起重要作用,并为临床治疗和预后判断提供一定的依据。
Objective It is to approach the expression of tumor suppressor gene PTEN protein in human gallbladder carcinoma tissues and its correlation with angiogenesis and other clinical biological behaviors.Methods The expression of PTEN protein in 37 cases of human gallbladder carcinoma and their adjacent tissue was detected by immunohistochemical staining.The microvessel density(MVD) was calculated by detecting antigen of CD34.Results The positive rate of PTEN in the gallbladder carcinoma was significantly lower than that in the adjacent normal tissue(P0.01).MVD of the gallbladder carcinoma was higher than that of the normal tissue(t=-7.94,P0.01).The expression of PTEN was significantly associated with histological differentiation(t=3.275,P0.01).As for the MVD of gallbladder carcinoma,there was significant difference between patients with lymph node metastasis and those without(t=-2.931,P0.01).Conclusion Negative expression of PTEN protein in gallbladder carcinoma is observed,which indicates that PTEN gene inactivation may be related to angiogenesis in gallbladder carcinoma,PTEN may play an important role in the genesis and development of gallbladder carcinoma.
