摘要
研究了牛血清白蛋白(BSA)在不同条件下荧光发射光谱的稳定性。结果表明,Tris-HCl、PBS和PB等缓冲液种类,常量和微量等石英比色皿类型,对于BSA的荧光稳定性没有明显影响。激发波长260 nm与280 nm对于BSA的荧光稳定性有显著性差异。超声和金属离子的干扰,都会导致BSA荧光强度的降低,在5%的误差范围内,各金属离子允许存在的倍数分别为:Cu2+(8),Zn2+(40),Mg2+(96),Ca2+(120)。本研究对于蛋白质溶液的制备和前处理具有一定的理论和实际意义。
Fluorescence stability of bovine serum albumin (BSA) solutions was investigated under differ ent conditions. The results indicated that buffer solutions, e. g. Tris-HCl, PBS and PB, and quartz cuvettes such as macro- and micro-cuvette did not affect fluorescence stability of BSA. BSA fluorescence showed significant stability diversity when excitation wavelength of 260 nm and 280 nm were used. Fluorescence intensity of BSA may be lowered by ultrasound or metal ions. Within 5% tolerance range of the detection value, the maximum permissible multiples of Cu2 ~ , Zn2 ~ , Mg2 ~ and Ca2 ~ were 8,40,96 and 120, respec- tively. This study was of certain theoretic and practical significance for preparation and pretreatment of protein solution.
