以GFP为报告基因优化农杆菌介导海岛棉转基因体系的研究 被引量:2

摘要 为了进一步建立优化的根癌农杆菌介导海岛棉外源基因转化体系,提高海岛棉外源基因转化率。以生理状态基本一致的海岛棉新海30号的胚性愈伤组织为受体材料,以绿色荧光蛋白基因(GFP)为报告基因,将浸染时间和共培养时间作为试验要素,筛选适宜的浸染时间和共培养时间。结果表明,根癌农杆菌介导的海岛棉外源基因转化体系的最佳浸泡时间为15 min,共培养时间为24 h。 This paper aims to ulteriorly establish an optimized agrobacterium-mediated transgenic system to raise transformation efficiency of exogenous gene in Gossypium barbadense.The Embryogenic callus of Xinhai 30 with the same physiological state and GFP gene respectively were used as recept materials and report gene,and the time of soak and co-cultivate were treated as experimental factors.The appropriate time of soak and co-cultivation were screened.The results indicate that the most suitable time of soak and co-culivation for agrobacterium-mediated transgenic system of Gossypium barbadense was 15min and 24h,respectively.
