摘要 目的了解杜仲总黄酮对新生大鼠颅盖骨体外培养成骨细胞骨钙素的mRNA表达的影响。方法杜仲总黄酮以10 g.mL-1、100 g.mL-1、200 g.mL-1浓度分别加入2d的SD大鼠颅骨分离培养的成骨细胞中,用Realtime-PCR法检测对骨钙素mRNA的表达。结果与对照组相比,各实验浓度的杜仲总黄酮均能促进骨钙素mRNA的合成。结论杜仲总黄酮成分能直接促进体外成骨细胞中骨钙素mRNA的表达。 Objective To approach the effects of total flavonoids of Eucommia Bark on the synthesis of Bone Gla Protein mRNA.Methods The osteoblast were isolated from the crinial bone of Sprague-Dawley rats at postnatal day 2,and cultured with 10μg·mL-1,100μg·mL-1and 200μg·mL-1 total flavonoids of Eucommia Bark.The mRNA of Bone Gla Protein was identificated by Real Time-PCR.Results The total flavonoids of Eucommia Bark at the concentration from 10μg·mL-1 to 200μg·mL-1 improved the synthesis of mRNA by osteoblasts.Conclusions The flavonoids of Eucommia Bark are the useful components promoting the synthesis of mRNA of Bone Gla Protein.
