乌鳖颗粒拮抗D半乳糖致小鼠卵巢早衰的实验研究 被引量:1

摘要 目的:探讨D半乳糖致雌性小鼠卵巢早衰动物模型的可行性和乌鳖颗粒的作用机理。方法:选取有正常动情周期的雌性小鼠,随机分为7组,每组10只,分别为正常组、模型组、己烯雌酚组、六味地黄丸组、乌鳖颗粒大、中、小剂量组。除正常组外,其余各组按每天0.5mL/只的剂量颈背部皮下注射5%D半乳糖溶液。第11天开始,灌胃给予治疗药,连续给药32天。每天定时观察阴道细胞,确定动情周期变化。实验末取血清测E2血清水平,取子宫和卵巢称湿重,计算子宫指数和卵巢指数,卵巢、子宫作组织形态学观察。结果:模型组小鼠动情周期紊乱,动情间期延长,乌鳖颗粒各剂量组小鼠在给药2周左右出现动情期延长。乌鳖颗粒各剂量组的卵巢指数、子宫指数与模型组小鼠比较具有显著性差异(P<0.01)。模型组小鼠的血清E2水平明显下降,与正常组比较具有显著性差异(P<0.01)。乌鳖颗粒大、中剂量组血清E2水平比模型组有明显的升高,与模型组比较具有显著性差异(P<0.05,P<0.01)。乌鳖颗粒各剂量组的卵巢、子宫组织形态学明显改善。结论:乌鳖颗粒能促进子宫、卵巢的生长发育,改善卵巢、子宫的组织形态,调节卵巢内分泌的激素水平。 Objective:Explore the feasibility of D-galactose to premature ovarian failure mice and the mechanism of Wu Bie granules.Methods:Selection normal oestrous cycle of female mice which were divided into seven groups of 10 each randomly,including,normal group model group,diethylstilbestrol group,Liu Wei Di Huang Group,Wu Bie granules of large,medium,low dose group.In addition to the normal group,and the remaining mice were given 5% D-galactose with 0.5 ml each day cervicodorsal subcutaneous injection.From the eleventh day,all mice were given the curative by ig.To determine the oestrous cycle by observing vaginal cells Every timing.At the end of the experiment,taking the serum and testing the level of E2.Take and weight uterus and ovary.Then calculation ovaries and uterine index.Ovarian for organization morphological observation.Results:Oestrous cycle of model control normal group mice were dysfunction and anoestrum were prolongation.Estrum of mice which had been given the corresponding medicine in two weeks were prolongation.Compared with model control group,ovarian index and uterine index of each dose group of Wu Bie particles were existing difference significantly.The level of serum E2 of high-dose of Wu Bie granules group and moderate doses of Wu Bie granules group increased obviously comparing with model control group existing difference significantly.The morphology of ovarian tissue of every dose of Wu Bie granules group improved obviously.The morphology of ovarian tissue of every dose of Wu Bie granules group improved obviously.Conclusion:Wu Bie granules can promote the development of uterus and ovary,improve The morphology of ovarian tissue,adjust ovarian hormones.
