宣痹汤对实验性IgA肾病大鼠肾脏MCP-1表达的影响

Effects of Xuanbi Decoction on Renal Pathology and Expression of MCP-1 in Renal Tissue in Rat Experimental IgA Nephropathy Model

摘要 目的:观察宣痹汤对实验性IgA肾病(IgA nephropathy IgAN)大鼠肾脏病理及肾组织单核细胞趋化因子(monocyte chemoattractive peptide1,MCP-1)表达的影响,了解其作用机制。方法:建立IgAN大鼠模型,随机分成正常对照组、模型组、雷公藤多苷组、宣痹汤低剂量组、宣痹汤中剂量组、宣痹汤高剂量组。用免疫组化和RT-PCR方法分别检测肾组织MCP-1蛋白和基因水平的表达。结果:与模型组相比,宣痹汤各治疗组对降低大鼠尿红细胞较雷公藤多苷治疗组明显(P<0.05);宣痹汤在基因和蛋白水平均能够有效抑制MCP-1在肾组织的表达(P<0.05)。结论:宣痹汤能降低IgA肾病大鼠尿红细胞和尿蛋白,下调MCP-1在肾脏的表达,这可能是宣痹汤治疗IgA肾病的作用机制之一。 Objective:To observe the effects of Xuanbi Decoction on renal pathology and expression of monocyte chemoattractive peptide1(MCP-1)in rat experimental IgA nephropathy model renal tissue at level of gene and protein,it can help to understand mechanism of action.Methods:Firstly,we constructed IgA nephropathy model,then divided models into the normal control group,the operation model group,triptergium wilfordi tablets group,Xuanbi Decoction low dose group,Xuanbi Decoction median dose group and Xuanbi Decoction high dose group.The expression of MCP-1 at the level of gene and protein in renal tissue were tested by immunohistochemistry and RT-PCR methods.Results:The Xuanbi Decoction group had more significant function of reducing the urine RBC than triptergium wilfordi tablets group;The Xuanbi Decoction could inhibit the expression of MCP-1 at the level of gene and protein in renal tissue.Conclusion:The Xuanbi Decoction can reduce the microscopic hematuria and proteinuria,downregulate the expression of MCP-1 in kidney,which may explain one of the mechanisms of Xuanbi Decoction effectiveness in clinical treatment of IgA nephoropathy.
