痛风合剂及其优化方对大鼠急性痛风性关节炎的疗效观察 被引量:9

摘要 目的:观察痛风合剂及其优化组方对尿酸盐所致急性痛风性关节炎大鼠的疗效,明确痛风合剂优化方的疗效是否更确切。方法:选取wista大鼠54只,随机分为9组。正常对照组、模型组、西药组、痛风合剂高、中、低浓度组、痛风合剂优化方高、中、低浓度组,分别用生理盐水、吲哚美辛混悬剂、痛风合剂高、中、低浓度汤剂、痛风合剂优化方高、中、低浓度汤剂灌胃7天,早晚各1次。于灌胃第四天把25g/mL(0.05mL)浓度的尿酸盐溶液注射到大鼠踝关节腔内,制备痛风性关节炎模型。对照组大鼠关节腔注射0.05mL生理盐水。分别于灌胃前和造模前、造模后24、48、72h秤取大鼠体重;造模前一天上午和下午,造模后0、2、4、6、8、10、12、24、48、72h测取右侧距小腿关节同一部位周径(取均值);造模后72h取血,观察血浆白细胞计数,检测血清白细胞介素-1β(Inter-leukin-1β,IL-1β)的含量;取血后将大鼠处死,取踝关节,10%福尔马林固定待做病理。结果:54只大鼠均进入结果分析。灌胃前体重无差异,造模后第1、2天痛风合剂及优化方高浓度组、西药组高于模型组,存在差异;造模前一天大鼠上午和下午踝关节周径无差异,造模后大鼠关节肿胀度各组均有所增高,其中以模型组最为明显,药物治疗后,用药组肿胀度均下降低,高浓度组和吲哚美辛效果相当;痛风合剂及优化方各剂量组血浆白细胞计数和血清白细胞介素1β水平明显高于对照组且以模型组最为显著,与模型组相比,痛风合剂及优化方高浓度组差异明显;病理组织学结果显示,痛风合剂及优化方可减轻痛风性关节大鼠踝关节的组织水肿和炎性细胞浸润,改善滑膜增生,且浓度越高作用越明显。但以上检测指标中,痛风合剂组和优化方组无差异。结论:痛风合剂及优化方可以有效抑制急性痛风性关节炎发作,且该作用呈一定的剂量依赖性,但该作用在痛风合剂和优化方之间并无差异。 Objective: To study the effect of gout mixture and its optimal mixture on rats acute gouty arthritis induced by monosodium urate,and whether the optimal mixture showed more superior effect than gout mixture.Method: 54 wista rats were randomly divided into nine groups.Control group,model group,western medicine group,gout mixture(highdoes,medium does,low dose) group,optimal mixture(high does,medium deos,low dose) group respectively were administrated of normal saline,indomethacin suspension,gout mixture of high,medium and low concentration,optimal mixture of high,medium and low concentration for 7 days,twice a day.rats acute gouty arthritis modles were estalished On the fourth day by monosodium urate solution(concentration of 25mg/mL,0.05mL) injected into the ankle cavity,while 0.05 mL normal saline was injected into joint cavity of rats in control group.The body mass was detected before administration of drug,model estabilishment and day 1,2,3 after modle estabilishment.The perimeter(mean value) of right jiont was measured in the same position at desigenated time(Am and Pm in the same day before model estabilishment and 0 2,4,6,8,10,12,24,48,72 hours after model estabilishment).After 72 hours of model estabilishment,blood samples were collected from inner canthus to count leukocyte and detect level of serum interleukin-1β;Executing rats after collecting blood,ankles were fixed to do Histopathologic with 10% formalin.Result:Totally 54 rats were involved in the result analysis.The body weights increased ongoingly before administation of drugs.After modle estabilishment,the increase of body mass in high dose of gout mixture and optimal mixture group and wester medicine group were significantly higher than other groups;Before model estabilishment,the perimete was no significant difference between AM and PM in one day.After model estabilishment,the ankles of rats swelled,especially in model group.After drug treatment,the swelling level was decreased.The high concentration of gout mixture and optimal mixture group were signifianctly difference from model groups,but compare to wester medicine group,there is no difference;Numeration of leukocyte and level of IL-1β in model group,gout mixture and optimal mixture group were higher than that of control model,especially model group.Gout mixture and optimization mixture of high dose group were lower than model group;The Histopathologic results showed that gout mixture and optimal mixture might reduce gouty joint edema,improve inflammatory cells infiltrating and synovial hyperplasia,the higher concentration of gout mixture and optimize mixture,the better of therapeutic effect.But from above indexes,gout mixture and optimize mixture group had no difference.Conclusion: Gouty mixture and optimal mixture can effectively inhibit the acute gout arthritis of rats,and the curative effect is dose dependent,but the effect is no difference between gout mixture and optimal mixture.
