摘要
优化达卡巴嗪脂质体的制备工艺并对其形态及体外释放进行研究。根据Box-Behnken的中心组合实验设计原理设计实验,以包封率、外观形态、体外释放为指标优化脂质体制备工艺。脂质体包封率为94.20%,比预测高0.02%,脂质体粒径在20~50μm。制得的脂质体的体外释放具有突释和缓释特点。预测工艺条件与实际工艺条件之间具有较好的拟合度,Box-Behnken中心组合设计可以应用于达卡巴嗪脂质体工艺条件的筛选。
To optimize the DTIC liposome preparation process and study the shape and the properties,we have the experiments according to Box-Behnken central composite principle,with indexes of the encapsulation efficiency,appearance,in vitro release.The Results show that Liposome encapsulation rate is 94.20% which is 0.02% higher than the predicted and the particle size is in 20~50 μm.We come to a conclusion that there are both the burst release and sustained-release in the Liposome,the predicted process conditions is similar to that of the real.The Box-Behnken central composite design can be applied to DTIC liposomes preparation.
