摘要
目的进一步探讨食管癌变与核基质蛋白的关系。方法对食管癌EC9706细胞行诱导分化及环六亚甲基双乙酰胺(HMBA)处理,处理前后双向电泳技术分析检测人食管癌EC9706细胞核基质蛋白表达变化。结果 HMBA处理后核基质蛋白15个蛋白点表达发生显著变化。其中表达明显减弱3种,表达明显增强7种,新出现5种。结论人食管癌EC9706细胞诱导分化过程中伴有核基质蛋白表达的差异,与癌细胞分化和逆转密切相关。
Objective To investigate further the relationship between esophageal carcinogenesis and nuclear matrix proteins. Methods Through cell culture and induction of differentiation treatment, nuclear matrix protein selectively extraction, two dimensional gel electrophoresis analysis was used to detect the expression of nuclear matrix protein in human esophageal carcinoma EC9706 before and after induced by hexamethylene bisacetamidc (HMBA)of 5 mmoL/L. Results There were 15 protein spots changed remarkably during the apoptosis induced by 5 mmol/L HMBA, 7 of which up-regulated, 3 of which down-regulated, and 5 were new spots. Conclusions There are significant changes of nuclear matrix pro- tein expression during the induction of human esophageal cancer EC9706 cells by HMBA.
