摘要
目的研究葛根素对实验性动脉粥样硬化兔炎症因子的影响,探讨其抗动脉粥样硬化的机制。方法雄性日本大耳白兔32只,随机分为正常对照组,模型组,葛根素低及高剂量组;正常组饲喂普通饲料,模型组在普通饲料中加入1%胆固醇及0.02%蛋氨酸,葛根素低及高剂量组高脂饲料喂养的同时给予葛根素(每天100 mg/kg,200 mg/kg),饲喂9 w,麻醉后采血,取主动脉上段组织-80℃保存。全自动生化仪测定各组血清总胆固醇(TC),低密度脂蛋白胆固醇(LDL-C);ELISA方法检测各组血清C反应蛋白(CRP),细胞间黏附分子-1(ICAM-1);HE染色观察主动脉病理学改变。结果与正常对照组相比,模型组血清TC,LDL-C,CRP,ICAM-1明显升高(P<0.01,P<0.05);与模型组相比,葛根素组血清TC,LDL-C,CRP,ICAM-1明显降低(P<0.01或P<0.05);病理学检验结果显示葛根素组动脉粥样硬化病变进展明显延缓。结论葛根素有抗动脉粥样硬化作用,该作用与抑制动脉粥样硬化过程中的炎症反应密切相关。
Objective To evaluate the effects of icariin on ability to prevent endothelial dysfunction in hypercholesterolemia rabbit.Methods Atherosclerosis was induced in rabbits by maintaining them on a hyperlipidaemic diet.24 Japanese male white rabbits were randomly divided into 3 groups with 8 rabbits each: the control group(C),the model group(M),the icariin group(ICA).Control group rabbits were fed with normal diet,and the other groups rabbits were fed with 1% cholesterol and 0.02% Methionine high lipid diet for 9 weeks.The icariin group received same diets plus in icariin(80 mg.kg-1.d-1 gastric gavage).Samples were collected and stored at-80 ℃ before assay.The serum levels of total cholesterol(TC) and low-density lipoprotein cholesterol(LDL-C) were measured by enzymatic assays,and C-reactive protein(CRP),intercellular adhesion molecule-1(ICAM-1) were measured with highly sensitive ELISA kit.The histological changes were observed by microscope with HE staining.Results Histopathological scores showed icariin had inhibitory effect on the progression of atherosclerosis.The level of TC,LDL,and CRP in model group increased significantly compared with control group(P 0.01 or P 0.05) and there was a trend towards increased serum level of ICAM-1 compared with control group(P 0.05).The level of TC,LDL and CRP in ICA group decreased significantly compared with model group(P 0.01 or P 0.05) and there was a trend towards decreased serum level of ICAM-1 compared with model group(P 0.05).Conclusions ICA treatment may have protective effect on endothelial dysfunction in hypercholesterolemia rabbit some extent.
