当归补血汤对高糖条件下系膜细胞增殖及乙酰肝素酶表达的影响 被引量:6

Effects of Astragalus and Angelica Mixture on the Proliferation of Glomerular Mesangial Cells and Expression of Heparanase in High Glucose Condition

摘要 目的:研究当归补血汤对高糖条件下肾小球系膜细胞(GMC)增殖及其乙酰肝素酶(HPA)表达的影响,探讨其在糖尿病肾病(DN)防治中的意义。方法:①利用透射电镜观察各组中GMC的增殖情况;②采用免疫组织化学方法检测各组肾小球中HPA的表达。结果:高糖能明显促进肾小球GMC增殖,也相应的增加系膜细胞中HPA表达(P<0.05);而当归补血汤能够明显抑制GMC增殖和相应减少HPA在肾小球中的表达(P<0.05),且效果优于格列奇特组。结论:高糖可以加强GMC增殖及细胞中HPA表达,当归补血汤能够明显抑制GMC增殖及细胞中HPA表达,从而预防肾小球硬化的发生、发展和恶化,进而延缓DN的进展。 Objective:To observe the effects of astragalus and angelica mixture on glomerular mesangial cells(GMCs) and the expression of heparanase(HPA) in high glucose condition,and to explore its significance of the prevention and treatment in the process of diabetic nephropathy(DN).Methods:Transmission electron microscopy was adopted to observe the proliferation of GMCs.The expression of HPA was determined by immunohistochemistry in glomerular in each group.Results:High glucose could promote the proliferation of glomerular GMCs,and the expression of HPA increased in mesangial cells(P0.05).The astragalus and angelica mixture inhibited the proliferation of GMCs and expression of HPA(P0.05),and the inhibitory effect was better than that of glibenclamide group.Conclusion:High glucose can enhance the expression of HPA and GMC proliferation,the astragalus and angelica mixture can inhibit the GMC proliferation and the expression of HPA,thus it may prevent the occurrence,development and deterioration of glomerular sclerosis,and delay the progress of DN.
