摘要
目的:旨在探讨黄芩素(BAI)对血管生成的影响,及血管内皮生长因子(VEGF)对多发性骨髓瘤细胞系RPMI-8226细胞生长的影响。方法:体外培养RPMI-8226细胞系,不同浓度BAI处理此细胞系,用Annexin-V/PI双标流式细胞术检测BAI对RPMI-8226细胞系凋亡的作用,应用RT-PCR检测黄芩素对RPMI-8226细胞VEGF表达的改变。结果:经不同浓度BAI(0,25,50,100μg/L)处理后的RPMI-8226细胞,随着BAI浓度的增加,细胞的早期凋亡及总凋亡率增加,以早期凋亡更明显;随着BAI(0,50,100μg/L)浓度增加,VEGF基因的表达呈下降趋势。结论:在一定浓度范围内,黄芩素可促进RPMI-8226细胞发生凋亡,且呈剂量依赖性,该过程与降低骨髓瘤细胞VEGF表达,抑制血管内皮生长作用有关。
Objective:To explore the effect of Baicalein(BAI) on angiogenesis and expression of vascular endothelial growth factor(VEGF) in multiple myeloma cell line RPMI-8226.Methods:Multiple myeloma cell line RPMI-8226 was cultured in vitro with different concentrations of BAI.Flow cytometry Annexin-V/PI double staining was used to evaluate the apoptosis effect of BAI on RPMI-8226 cells,and the transcription level changes of VEGF gene were determined by RT-PCR.Results:Flow cytometry Annexin-V/PI double staining revealed that early apoptosis and total apoptosis rate of RPMI-8226 cells increased as BAI concentration(0,25,50,and 100μg/L) increased.RT-PCR detection of VEGF gene showed that the trend of its transcription level was increasing according to the increasing of BAI concentration(0,50,and 100 μg/L).Conclusion:Baicalein can induce the apoptosis of RPMI-8226 cells within a certain range of concentration in a dose-dependent manner,maybe through decreasing the expression of VEGF and inhibiting the growth of vascular endothelium.
