Effect of granulocyte-colony stimulating factor on the differentiation of implanted human neural stem cells in vivo

摘要 目的观察缺氧缺血性脑损伤(HIBD)新生大鼠应用粒细胞集落刺激因子(G-CSF)对移植的人神经干细胞(hNSCs)体内分化成星形胶质细胞的影响。方法新生1周的Wistar大鼠建立HIBD模型后分成2组:NSC移植组(A组,n=16)和NSC移植+G-CSF治疗组(B组,n=16),其中B组建模后1 h即开始皮下注射G-CSF,1次/d,移植前后连续5 d注射G-CSF。两组均于建模后第2天经脑室移植hNSCs。移植后1、2周分别用免疫荧光法检测植入细胞在大鼠脑内向星形胶质细胞分化的情况。结果移植后1周,B组植入细胞分化的星形胶质细胞数量较A组多,差异有统计学意义(t=4.83,P<0.01)。移植后2周,两组植入细胞分化的星形胶质细胞数量上差异无统计学意义(t=1.59,P>0.05)。结论 G-CSF能在移植后早期促进植入的外源性NSCs向星形胶质细胞分化。 Objective To examine the effect of granulocyte-colony stimulating factor(G-CSF)on the differentiation of implanted human neural stem cells(hNSCs)into astrocytes in neonatal rats with hypoxic-ischemic brain damage(HIBD).Methods One week old rats with HIBD were divided into two groups:the hNSCs transplantation without treatment group(Group A,n = 16)and the hNSCs transplantation with G-CSF treatment group(Group B,n = 16).G-CSF(50 μg/(kg·d))was injected subcutaneously one hour after operation initially,and then once a day for five days in a row.In both groups,animals were implanted with hNSCs at the left cerebral ventricle on the second day following operation.The astroglial differentiation of the transplanted cells in rat brains was detected by immunofluorescence one week and two weeks after the transplantation.Results Differentiated astrocytes from the implanted cells distributed mainly in the corpus striatum,white matter and cortex on the injured side.The number of differentiated astrocytes was greater in the rats of group B than that in group A one week post transplantation(t = 4.83,P 0.01),while no differences exited another one week later(t = 1.59,P 0.05).Conclusions G-CSF promotes the differentiation of implanted hNSCs into astrocytes in the early stage.
