摘要
采用试纸片法和肉汤稀释法测定重组溶葡球菌酶(rLspn)的抑菌圈大小、最小抑菌浓度(MIC)和最小杀菌浓度(MBC),通过与溶葡球菌酶标准品和两种抗生素相比较,评价rLspn的体外抑菌活性。试纸片法试验结果显示,rLspn有专一抑制金黄色葡萄球菌的作用,而对其他菌株无效;rLspn与标准品的抑菌活性差异不显著(P>0.05);对于耐青霉素的金黄色葡萄球菌,rLspn仍然表现出良好的抑菌活性。肉汤稀释法结果表明rLspn的抑菌活性优于对耐药金黄色葡萄球菌最有效的盐酸万古霉素,MIC为0.1μg/mL^0.2μg/mL。
The size of inhibition zone,the minimun inhibitory concentration(MIC) and the minimal bactericidal concentration(MBC) of the recombinant lysostaphin rLspn were assessed by disc agar diffusion test and broth dilution test,through comparing with lysostaphin and two kinds of antibiotics,to appraise the bacteriostasis activity of recombinant lysostaphin in vitro.Disc agar diffusion test showed that rLspn suppressed Staphylococcus specificallx,but was invalid to other strains.The bacteriostasis activity of rLspn and Lspn was not significant difference(P0.05).Regarding the Staphylococcus resisting to penicillin,rLspn still showed good bacteriostasis activity.Broth dilution test indicated that the bacteriostasis activity of the rLspn was higher than that of hydrochloric acid vancomycin,which was effective to antibiotic resi-stence Staphylococcus,and the MIC was 0.1 μg/mL-0.2 μg/mL.
