摘要
应用PCR快速检测急性病死猪的病原。采集病死猪脾等组织,用PCR方法检测猪繁殖与呼吸综合征病毒(PRRSV)、猪瘟病毒(CSFV)和猪圆环病毒2型(PCV-2)。同时采集病死猪肝、心血,无菌接种血平板进行细菌分离,用16 S rRNA基因测序法鉴定分离菌。对分离菌株进行药敏试验。PCR检测结果显示检测样品用CSFV引物扩增出475 bp的条带,用PCV-2和PRRSV引物未扩增出条带。细菌分离结果显示,在血平板出现针尖大小的菌落,16 S rRNA基因测序法鉴定分离菌为猪丹毒杆菌。药敏试验结果显示,分离菌对强力霉素和环丙沙星等药物敏感。应用PCR方法对病死猪的病原快速鉴定,结果表明病死猪为猪瘟病毒和猪丹毒杆菌混合感染。
The pathogens that infected swine were diagnosed with PCR.Samples from spleen of swine were detected for Porcine reproductive and respiratory syndrome virus(PRRSV),Classical swine fever virus(CSFV),and Porcine circovirus type 2(PCV-2),by using PCR.In addition,bacteria were isolated and cultured with blood plates from liver and blood of diseased swine,and identified with sequence analysis of 16 S rRNA,and the medicine sensitivity was studied.The result showed that a 475 bp fragment could be amplified with CSFV primier,and PCV-2 and PRRSV were detected from samples.The pinpoint-shaped colonies were taken and isolated with blood plates,and sequence analysis of 16 S rRNA showed that it was a strain of Erysipelothrix rhusiopathiae.The strain was sensitive to doxycycline,ciprofloxacin,et al.These results showed that the sick and dead swine were diagnosed as Classical swine fever virus and Erysipelas suis coinfection.
