摘要
本文采用离体家兔心脏制备心肌细胞膜,观察了ATP-MgCl_2,和ATP对缺血缺氧心肌细胞膜钠钾-ATP酶活性的影响及活性变化规律,探讨了ATP-MgCl_2,对心肌的保护机制。实验中发现,ATP-MgCl_2,对缺血缺氧心肌的保护作用优于ATP,家兔心脏缺氧5,10,20,30 min时,其心肌细胞膜钠钾-ATP酶活性分别下降至59.57%,44.64%,23.66%,9.96%,缺氧60min时,其活性完全消失,给予ATP-MgCl_2以后,除了60 min组以外,各缺血组的钠钾-ATP酶活性都有显著的提高,缺血60 min组其活性未见恢复,给予ATP以后钠钾-ATP酶活性也有一定的提高,但其效果不明显。ATP-MsCl_2组与ATP组之间有显著性差异。
Using the myocardial cells isolated from rabbit heart, the effect of ATP on Na^+-K^+-ATPase activity in the plasma membrane of anoxemic myocardial cell was observed. The results were as follows: The cytoprotective effect of ATP-mgCl_2 on anoxemic myocardial cell was superior to that of ATP. At 5, 10, 20, 30 minutes after anoxemia, the Na^+-K^+-ATPase activity decreased to 59.57, 44.64, 23.66 and 9.96% of control respectively. Administration of ATP-MgCl_2 could increase the ATPase activity (P<0.01), but ATP itself did not have the effect (P<0.05). The results mentioned above suggest that high-energy phosphate has significant cytoprotective effect on anoxemic myocadial cells and the effect of ATP-MgCl_2 is superior to that of ATP. The cytoprotective effect of ATP-MgCl_2 may be partly due to its effect of increasing Na^+-K^+-ATPase activity.
