NMDA受体亚单位NR1在离体人神经干细胞中的表达

摘要 目的研究离体培养的人海马神经干细胞(NSCs)中NMDA受体亚单位NR1的表达。方法分离培养胎龄8-12周人胚脑海马神经干细胞,对NSCs进行Nestin和分化鉴定。用免疫细胞化学、Western blot免疫印迹和RT—PCR等方法检测原代、传代1次、传代2次的人胚胎海马NSCs中NR1蛋白和mRNA的表达。结果在孕8~12周人胚脑海马分离培养的NSCs中,NMDA受体亚单位NRI免疫细胞化学反应呈阳性,该受体亚单位的蛋白和mRNA均被检测钊。结论体外培养的早期人胚胎海马NSCs能稳定表达NMDA受体亚单位NR1。 Objective To investigate the expression of N - methyl - D - aspartate (NMDA) receptor subunit 1 in the cultured human hippoeampus neural stem cells (NSCs). Methods NSCs from hippocampus of abortion human embryonic tissue (8 -12 weeks post conception) were cultured and passaged in suspension. The nemospheres were identified by Nestin immunocytochemistry and the differentiation potential of NSCs were also detected. The protein and mRNA expression of NMDA receptor subunlt 1 in the human hippocampus NSCs of primitive,passage 1 and 2 were measured by immunocytochemistry, Western blot and reverse transcription polymcrase chain reaction ( RT - PCR). Results NR1 immunostaining positive cells could be found in the human hippocampus NSCs. The protein and mRNA of NR1 were identified in the same cells. Conclusion NSCs from the human embryonle bippoeampus can express NMDA receptor subunitl in vitro.
