摘要
n-3多不饱和脂肪酸脱氢酶基因fat-1来自于秀丽线虫(C.elegans)。为检测该基因在人肺癌细胞H460中的表达效果,本项研究构建了哺乳动物表达载体pcDNA3.1(+)myc-HisA-fat-1,以Xfet polymer介导法转染到人肺癌细胞H460中,RT-PCR检测到有效的异源基因表达,MTT法证实基因表达能有效地抑制肺癌细胞的增殖率(P<0.05),气相色谱分析基因表达前后细胞中n-6/n-3多不饱和脂肪酸比例降低(P<0.05),为将该基因用于癌症的转基因治疗奠定了基础。
fat - 1 gene is a kind of n - 3 fatty acid dehydrogenase gene from Caenorhabditis elegans. In this study, the eukaryotic expression vector pcDNA3.1 ( + ) myc - HisA - fat - 1 was constructed and expressed in human lung cancer cell H460. RT - PCR results showed that fat - 1 gene could expressed effectively in H460 cell and cell proliferation rate was markedly inhibited ( P 〈 0.05 ). Moreover, fat - 1 gene could significantly decreased the ratio of cellular n - 6/n - 3 PUFAs I P 〈 0.05 ).
