摘要 为了解辐射后蝴蝶兰的遗传情况,本试验以60Co-γ射线为辐射源,采用梯度剂量率(0、15、20、25和30 Gy)对蝴蝶兰F141780进行处理。以诱变苗为母体,取其花梗腋芽作为外植体进行培养。取组培苗新叶提取g-DNA,运用RAPD-PCR技术进行扩增。将组培苗与诱变苗在基因层面上进行比对。结果表明,低剂量组同组材料间遗传距离较近,遗传的稳定性较强;高剂量组同组材料间遗传距离较远,遗传的稳定性较差。 In order to understand the heredity of radiated Phalaenopsis aphrodite,this research used graded doses(0,15,20,25 and 30 Gy) of radiation of ^60Co γ-ray as source to treat Phalaenopsis aphrodite cv.F141780,and then the axillary bud of floral stalk from mutant seedlings was taken as explants to cultivate.The genomic DNA of the cultivated tissue was extracted from young leaves and amplified using RAPD-PCR technique.The differences between the cultivated tissue from normal and mutant seedlings were compared at gene level.The ma-terials in the same group of low-dosage groups have a small genetic distance and strong genetic stability,while the materials in the same group of high-dosage groups have long genetic distance and weak genetic stability.
