摘要
目的探讨脂肪源性干细胞(ADSCs)移植对大鼠脑缺血再灌注损伤海马区乏氧诱导因子HIF-1α和HIF-3α表达的影响。方法线栓法制作大鼠大脑中动脉缺血(MCAO)2 h再灌注模型,18只大鼠随机分为假手术组(n=6)、模型组(n=6)、ADSCs组(n=6)。ADSCs组经尾静脉注射PKH-26标记的ADSCs细胞悬液(0.5 ml,细胞密度为4×106个/ml)。缺血3 h再灌注72 h后行NSS法行神经功能评分,Western blot和实时荧光定量PCR检测海马区HIF-1α和HIF-3α蛋白和mRNA表达。结果 ADSCs移植后可见PKH26标记的ADSCs在海马区有表达。与模型组比较,ADSCs组神经功能损害评分显著降低(P<0.05),海马区HIF-1α和HIF-3α蛋白和mRNA表达显著上调(P<0.05)。结论脂肪源性干细胞移植可上调脑缺血再灌注损伤大鼠海马区HIF-1α和HIF-3α表达,促进脑缺血损伤神经功能恢复。
Objective To investigate the effects of transplantation of adipose-derived stem cell(ADSC) on the expression of HIF-1α and HIF-3α in hippocampus region after cerebral ischemia-reperfusion in rats.Methods The rat model of middle cerebral artery occlusion was established by the suture method.Rats were randomly divided into sham operation group(n=6),model group(n=6) and ADSCs group(n=6).ADSCs suspension were marked with PKH-26 and then infused into the caudal vein in ADSCs group(0.5 ml,4×106/ml).After ischemia for 2 h and reperfusion for 72 h,neurological function was investigated by neurological severity scores(NSS),and the expression of HIF-1α and HIF-3α in hippocampus region was detected by Western blot and quantitative RT-PCR.Results After the transplantation of ADSCs,PKH-26-labeled ADSCs were observed in hippocampus region.Compared with model group,the neurological severity scores in ADSCs group were significantly decreased(P0.05),and the protein and mRNA expression of HIF-1α and HIF-3α in hippocampus region was significantly increased(P0.05).Conclusion ADSCs transplantation could promote the nerve functional recovery after cerebral ischemia injury by elevating the expression of HIF-1α and HIF-3α in hippocampus region.
