高浓度RA对拟胚体胚层分化的影响

摘要 目的 探讨维甲酸(RA)在拟胚体分化中对胚层分化的影响.方法 将胚胎干(ES)细胞悬浮培养形成胚胎小体(Embryonic bodies,EBs),用高浓度RA诱导,利用免疫荧光和RT-PCR方法比较RA对三胚层分化的影响.结果 高浓度RA诱导EBs中神经外胚层的基因和蛋白表达,抑制中胚层和内胚层的基因和蛋白表达.结论 RA在胚胎干细胞分化早期就决定了胚层的分化,高浓度RA促进神经胚层分化. Objective Discussion retinoic acid (P.A) in embryoid body differentiation of mesoderm differentiation. Methods To embryonic stem(ES) cell suspension culture to form embryonic bodies (Embryonic bodies, EBs), with high concentrations of RA-induced, using immunofluorescence and RT-PCR method is RA on differentiation of three germ layers. Results High concentrations of RA-induced EBs in neuroectodermal gene and protein expression, inhibition of mesoderm and endoderm gene and protein expression. Conclution RA in the early differentiation of embryonic stem cells determine the endoderm differentiation, high concentrations of RA to promote neural differentiation of mesoderm.
