目的:探讨创造酶切位点与PCR-RFLP原理在基因多态检测中的应用及其注意事项。方法:应用创造酶切位点原理设计引物,通过PCR-RFLP技术判断多个基因多态性。结果:共对MGMT、ADH2、MTH-FR和PON2基因多种多态位点进行了实验研究,均成功完成检测。结论:应用创造酶切位点PCR-RFLP原理建立的检测方法具备简便、经济、快速的特点,适宜推广应用。

Objective：To discuss attentions and application methods of create restriction site and PCR-RFLP to identify gene polymorphisms.Methods：Theory of restriction site was used to design primers,and PCR-RFLP technology was used to identify gene polymorphisms.Results：Polymorphisms of MGMT,ADH2,MTHFR and PON2 gene were detected successfully.Conclusion：It is suggested that method of create restriction site and PCR-RFLP for detecting polymorphism is facility,economy,and rapidness,and which should be widely applied.
