摘要
目的了解广东省外观健康犬狂犬病病毒带毒率情况。方法使用DFA法对随机采取的外观健康犬脑组织进行抗原筛查,再以RT-PCR和MIT法对DFA阳性样品进行复检,RT-PCR阳性的样品进行狂犬病毒N基因片段的扩增、测序和比较分析。结果在1 833份样品中,检出阳性样品2份,狂犬病病毒阳性率0.11%。序列分析显示这两株广东分离株属于狂犬病毒I型,基因同源性为99.7%。结论广东省外观健康犬中存在狂犬病毒感染情况,分离毒株与湖南和广西分离株有高度的亲缘关系。
In order to understand the infection rate of healthy appearance domestic dogs with rabies in Guangdong Province,brain samples from healthy appearance domestic dogs were collected and detected by direct immunofluorescence assay(DFA).Then the positive samples by DFA were further detected by nest RT-PCR and MIT.The positive samples detected by RT-PCR were sequenced with a part of N gene.There were two positive samples in 1833 and the positive rate was 0.11%.The sequences of N gene of two positives were identical,with the homogeny of 99.7% on nucleotide level,and they belonged to Lyssavirus genotype I.Thus dogs infected with rabies virus were found in Guangdong Province and this study showed that these virus had a close genetic relationship with the rabies virus which were found in Hunan and Guangxi provinces.
