针刺干预对脑缺血再灌注损伤大鼠外周血清蛋白表达的影响 被引量:1

Effect of Acupuncture on the Expression of Peripheral Serum Protein in Rats with Cerebral Ischemia Reperfusion Injury

摘要 目的:探讨脑缺血再灌注损伤模型大鼠外周血清蛋白表达的变化及针刺百会、足三里穴的干预作用。方法:以大脑中动脉线栓法制备大鼠脑缺血再灌注模型,采用随机区组设计将大鼠分为模型组、假手术组和针刺组3个大组,每大组再按照再灌注后时间分成12 h、24 h、48 h、72 h、96 h和144 h等6个亚组;每个亚组有大鼠7只;另设正常对照组7只。应用SELD I-TOF-MS检测各组大鼠外周血清蛋白表达的差异。结果:模型组与正常对照组比较,有150个差异蛋白峰(P<0.05);针刺组与模型组结果比较,有89个差异蛋白峰出现显著性变化,包括正向峰32个和负向峰57个;模型组血清蛋白峰值的分布呈双峰型,针刺干预使第一峰峰值降低,第二峰峰值时间前移。结论:针刺百会和足三里穴能够通过影响脑缺血再灌注损伤大鼠外周血清蛋白的表达,干预缺血再灌注模型大鼠的病理生理过程。 Objective:To explore the expression of peripheral serum protein expression in cerebral ischemia/reperfusion injury(CI/RI) rats and the influence of acupuncture points of Baihui(DU20) and Zusanli(ST36)on it.Methods:CI/RI model was established by using right middle cerebral artery occlusion and reperfusion.SD rats were randomly divided into model group,sham-operation group and acupuncture group,each group was further divided into 12 h,24 h,48 h,72 h,96 h and 144 h six subgroups(7 rats in each subgroup),adding 7 normal rats as control group.Surface enhanced laser desorption/ionization-time of flight-mass spectrometry(SELDI-TOF-MS) technology were used for inspecting the differences of peripheral serum protein expression in each group.Results:The result of SELDI-TOF-MS analysis showed that compared with control group,model group had 150 difference signal peaks related to CI/RI(P0.05),and there were 89 differences protein peaks in acupuncture group and model group,including 32 positive peaks and 57 negative peaks.The serum protein peaks present double-peak in model group,while the acupuncture intervention could lower the first protein peak,and made the second peak arriving ahead.Conclusion:Acupuncturing DU20 and ST36 can affect the expression of peripheral serum protein in CI/RI rats,interfering the pathophysiologic process of CI/RI.
