细胞间通讯与前列腺素合成——角膜内皮细胞与血小板相互作用合成前列环素和血栓素

摘要 体外培养的兔角膜内皮细胞具有合成前列环素(PGI_2)的能力,而不能合成血栓素(TXA_2)。与此相反,血小板本身能够合成TXA_2却几乎不能合成PGI_2。当二者共同培养时,前者可利用后者的前列腺素前体选择性地增加了PGI_2的合成,而后者却不能利用前者的前列腺素前体。这些发现动摇了角膜内皮细胞只是被动地参与眼前节炎性反应的一般概念,表明角膜内皮细胞与血小板之间经过细胞间“通讯”调节PGI_2和TXA_2的平衡,主动地参与眼前节的功能。 Prostaglandin-mediated intercellular communications between cultured rabbit corneal endothelial cells (RCE) and rabbit platelets (RP) were demonstrated. Prostacyclin (PGI2) and thromboxane A2(TXA2)were measured by radioimmunoassay. RCE perse synthesized PGI2 but not TXA2. In contrast, RP synthesized TXA2 but not PGI2. However in the combined RCE-RP culture, only RCE selectively utilized the precursor of prostaglandin, which is produced by RP to synthesize PGI2 but not vlce-versa. These findings may challenge the conventional concept suggesting corneal endothelial cells just passively be involved in inflammatory process of ocular anterior segment. Therefore, the experimental model for studying intercellular communication among corneal endothelial cells, vessel endothelial cells and platelets was posed.
