雷丸多糖的提取分离及其抗氧化活性研究 被引量:13

摘要 目的:研究雷丸多糖的提取、分离技术及其抗氧化活性。方法:用纤维素酶法酶解后对雷丸多糖进行提取工艺研究。通过Sevage法除蛋白,乙醇沉淀,活性炭脱色,再经Sephadex G-200柱层析法对其进行纯化,得到雷丸多糖OL1、OL2和OL3,并对其进行了抗氧化活性的研究。结果表明:酶解雷丸多糖的最佳提取工艺条件是:酶解温度40℃,酶添加量0.08%,酶解时间90min。结论:纤维素酶法可大大缩短雷丸多糖的提取时间,是极具应用前景的提取方法。雷丸多糖具有较强的清除自由基能力,是天然抗氧化剂的良好来源,可以进一步开发利用。 Objective:To investigate the extraction and purification of Omphalia lapidescens polysaccharide and its antioxidant activities.Methods:The O.lapidescens polysaccharide was extracted after enzymolysis by cellulase,through alcohol fractional precipitation,free proteins and colors were removed by Sevage method and charcoal,respectively.OL1、OL2 and OL3 were isolated and purified by Sephadex G-200 column.The best condition for enzymatic hydrolysis of O.lapidescens polysaccharide was studied:adding amount of cellulase 0.08%,enzymatic hydrolysis for 90 min at 35 ℃.Results:The extraction time was reduced greatly by cellulase enzymatic hydrolysis method,which indicates that it is a potential method for extraction polysacchatides from O.lapidescens.Omphalia polysaccharide is a kind of good source for natural antioxidants,has relatively strong antioxidant activities and worth further development and utilization.
