摘要
目的探讨Ghrelin对高脂环境下大鼠成肌细胞糖脂代谢的影响,为深入研究2型糖尿病的防治提供理论依据。方法对照组用含0.5%BSA的DMEM培养12 h;高脂组用0.3 mmol/L棕榈酸(PA,含0.5%BSA)培养12 h;干预组分为3组,分别在高脂干预同时加入10-11 mol/L、10-9 mol/L、10-7 mol/L Ghrelin作用12 h。用油红O染色法检测成肌细胞脂肪变性后甘油三酯沉积;用GPO-POD酶法测定成肌细胞脂肪变性后甘油三酯含量;用同位素示踪法检测葡萄糖摄取。结果与对照组比较,在0.3mmol/L PA组中,甘油三酯沉积和含量明显增加,葡萄糖摄取明显下降。与0.3 mmol/L PA组对比,随着给予Ghrelin的浓度逐渐增加,甘油三酯沉积和含量逐渐下降,葡萄糖摄取逐渐增加,并呈剂量依赖关系,其中10-7 mol/L Ghrelin对损伤的修复作用最为明显。结论 Ghrelin可减少大鼠成肌细胞的甘油三酯沉积,增加高脂环境下大鼠成肌细胞的糖摄取,并且存在剂量依赖关系。
Objective To investigate the effects of Ghrelin on glucose metabolism in myoblasts in rats in high-fat environment, and to provide theoretical evidence for studies on prevention and cure of type 2 diabetes. Methods The control group was cultured in DMEM with 0.5% BSA for 12 hours (h). The high-fat group was exposed to palmitic acid with the concentration of 0.3 mmol/L for 12 h. There were three intervention groups,in which the myoblasts of rats were exposed to 0.3 mmol/L palmitic acid for 12 h,at the same time, 10-11 mol/L, 10-9 mol/L, 10-7 mol/L Ghrehn were added,respectively. Then,the Oil-Red O dyeing method was used to display the TG sediment in cells. GPO-PODase method was used to determine the TG content in cells. And isotope tracer method was conducted to show the influence on 2- dexoy-D-[ 3H ]glucose uptake of myoblast. Results Compared with the control group, TG content and its sediment increased and [ 3H ]-G uptake decreased obviously in 0.3 mmol/L PA group. Compared with the 0.3 mmol/L PA group, TG content and its sediment decreased and [3H]-G uptake increased with the concentration of Ghrelin increased in the intervention groups,and the effect was most obvious with 10-7 mol/L Ghrehn. Conclusion Ghrelin can reduce the TG sediment and increase glucose-uptake in myoblasts of rats,and this effect is related with PA concentration.
